"PNAS" RNAi papers withdrawn

This paper was published in the journal PNAS in 2009 and identified an RNA-dependent RNA polymerase (RdRP) that plays a role in RNAi in Drosophila.

According to Concetta Lipardi and Bruce M. Paterson, authors of NCI, the D-elp1 subunit in the Drosophila RNA polymerase II extension complex is an atypical RdRP. The authors demonstrated the RdRP activity of D-elp1 by nearest neighbor analysis. In addition, they claim that the D-elp1 subunit plays a role in RNAi, transposon suppression and endogenous siRNA production.

For this paper, Luc Swevers, a scientist at the National Science Research Center of Demokritos in Greece, said that there were some doubts when the paper was received, but there are some important findings, such as the role in RNAi. "The presence of RdRP in insects will indicate that the RNAi reaction may be amplified."

However, on August 5 this year, PNAS withdrew this article on behalf of the author. Further investigation revealed that Lipardi and Paterson inadvertently misinterpreted the results of the nearest neighbor analysis experiment. Near-neighbor analysis actually determined the RNA template rather than the second RNA strand synthesis.

RNA-dependent RNA polymerase plays an important role in RNA silencing of several model organisms, such as Arabidopsis, Schizosaccharomyces pombe, and nematodes. In this process, RdRP uses RNA as a template to replicate RNA, expanding the gene silencing function of siRNA.

Although the researchers hypothesized that a similar RNAi mechanism exists in Drosophila, they have not found a similar protein in Drosophila. If this protein does exist in Drosophila, as Lipardi and Paterson said, then this RNAi mechanism may also exist in higher eukaryotes and even humans.

At the NCI Cancer Research Center, Paterson leads the biochemistry and gene expression department of the Biochemistry and Molecular Biology Laboratory. His research team is devoted to the gene regulation of eukaryotes, especially the RNAi of Drosophila. Lipardi previously worked under Peterson, but she recently joined the pharmaceutical manufacturer Merck.

The researchers apologize for their misunderstanding, but still emphasize that the D-elp1 subunit plays an important role in RNAi, endogenous siRNA production and transposon suppression, and their research on these effects will continue.
